The starting point for a typical route-scouting project was an 18-step discovery route. This sequence involved multiple chromatography steps and resulted in a low overall yield of the desired product.
The process route scouting project was handled in three distinct stages:
- Company-wide brainstorming: 23 new suggestions led to 4 alternative synthetic routes (1 FTE, 2 weeks)
- Route scouting: an optimized 6-step route on the gram scale, was found, including a key biocatalytic transamination (2 FTE, 6 weeks)
- Verification: successful scaling up of the key steps to the kilogram scale in a multiparameter optimization study
